UPLC指纹图谱技术结合毒性成分含量优选苍耳子的炮制工艺

摘要(Abstract):

通过16个不同工艺炮制样品水提取部位的UPLC指纹图谱和毒性成分含量比较,优选苍耳子炒制的最佳工艺。UPLC指纹图谱色谱条件为Acquity BEH C18色谱柱(2.1 mm×100 mm,1.7μm);流动相乙腈-0.1%磷酸溶液,梯度洗脱;流速0.25 mL·min-1;检测波长327 nm。毒性成分含量测定的色谱条件为Agilent TC-C18色谱柱(4.6 mm×250 mm,5μm);流动相甲醇-0.01 mol·L-1磷酸二氢钠溶液(35∶65);流速1.0 mL·min-1;检测波长203 nm。实验建立了16个不同工艺样品的UPLC指纹图谱,并对其分别采用《中药色谱指纹图谱相似度评价软件》、SPSS16.0软件和SIMCA13.0软件进行相似度分析、聚类分析和主成分分析;16个炮制样品相似度均大于0.97;聚类分析表明炮制样品可大致聚成4类;主成分分析结果认为影响最明显的3个成分分别是绿原酸、3,5-二咖啡酰奎宁酸和咖啡酸;毒性成分含量测定结果表明炮制后毒性成分明显降低,炮制可起到减毒作用;该实验方法可用于优选苍耳子的炮制工艺。
The experiment's aim was to optimize the processing technology of Xanthii Fructus which through comparing the difference of UPLC fingerprint and contents of toxicity ingredient in water extract of 16 batches of processed sample. The determination condition of UPLC chromatographic and contents of toxicity ingredient were as follows. UPLC chromatographic: ACQUITY BEH C18column( 2. 1 mm × 100 mm,1. 7 μm) eluted with the mobile phases of acetonitrile and 0. 1% phosphoric acidwater in gradient mode,the flow rate was 0. 25 mL·min-1and the detection wavelength was set at 327 nm. Contents of toxicity ingredient: Agilent TC-C18column( 4. 6 mm ×250 mm,5 μm),mobile phase was methanol-0. 01 mol·L-1sodium dihydrogen phosphate( 35∶ 65),flow rate was 1. 0 mL·min-1,and detection wavelength was 203 nm. The chromatographic fingerprints 16 batches of samples were analyzed in using the similarity evaluation system of chromatographic,fingerprint of traditional Chinese medicine,SPSS16. 0 and SIMCA13. 0 software,respectively. The similarity degrees of the 16 batches samples were more than 0. 97,all the samples were classified into four categories, and the PCA showed that the peak area of chlorogenic acid,3,5-dicaffeoylquinic acid and caffeic acid were significantly effect index in fingerprint of processed Xanthii Fructus sample. The outcome of determination showed that the toxicity ingredient contents of all samples reduced significantly after processing. This method can be used in optimizing the processing technology of Xanthii Fructus.
