玄参治疗大鼠内毒素血症的血清蛋白质组变化的初步研究

摘要(Abstract):

目的 :研究玄参治疗大鼠内毒素血症的血清蛋白质组变化。方法 :用双向电泳 (2DE)分析正常组、内毒素 (lipopolysaccharide ,LPS ;静脉注射 )组、玄参 (灌胃给药 )组、LPS(静脉注射 ) +玄参 (灌胃给药 )组之间的血清蛋白质组表达差异。结果 :①LPS致内毒素血症大鼠的血清在 2DE胶上 16个蛋白点 (xPr)出现非常明显的含量变化 ,与正常组比较 ,LPS组的xPr16 ,xPr19的点容量值均极显著降低 ,xPr1,xPr2 ,xPr3,xPr4 ,xPr5 ,xPr6 ,xPr7,xPr8,xPr9,xPr11,xPr12 ,xPr14 ,xPr18,xPr2 3的点容量值均极显著增高。②LPS +玄参组与LPS组比较 ,LPS引起增高的xPr1,xPr6 ,xPr7,xPr8,xPr9,xPr11,xPr12 ,xPr14 ,xPr18,xPr2 3的点容量值均显著降低。玄参在xPr1,xPr8,xPr11,xPr12 ,xPr18均表现对LPS显著的交互作用。LPS +玄参组与正常组比较 ,其中xPr8,xPr9,xPr11,xPr12 ,xPr14 ,xPr2 3蛋白点已被明显调节到正常状态 ,但是xPr18的点容量值显著低于正常组 ,xPr1,xPr6 ,xPr7的点容量值仍显著高于正常组。③玄参与LPS两因素对xPr5 ,xPr10 ,xPr19,xPr2 0 ,xPr2 1,xPr2 2均有明显的交互作用。④在玄参组 ,xPr13,xPr2 0 ,xPr2 1,xPr2 2的点容量值均比正常组显著增高 ,xPr10 ,xPr15的点容量值均比正常组显著降低 ,这 6个蛋白点在LPS组
Objective: To study the serum proteome of ra t endotoxemia treated by figwort root (FR). Method: The di fferences of serum proteome among rats treated with lipopolysaccharide(LPS), FR, LPS+FR and saline respectively were analyzed by two-dimentional electrophoresi s (2DE) assay. Result: The volumes of sixteen serum protei ns (xPr) in LPS induced-endotoxemia group were greatly changed compared with th ose of the control group. Among them, the volumes of xPr 16, 19 were significan tly decreased, and the volumes of xPr1, 2, 3, 4, 5, 6, 7, 8, 9, 11, 12, 14, 18, 23 were significantly increased. When treated with FR, the volumes of xPr1, 6, 7 , 8, 9, 11, 12, 14, 18, 23 were significantly decreased, and the volumes of xPr8 , 9, 11, 12, 23, 14 were back to normal level. Two factors statistic analysis sh owed that FR had interaction with LPS for xPr 1, 5, 8, 10, 11, 12, 18, 19, 20, 21, 22, and FR might be the functional antagonist of LPS. We also observed that the volumes of xPr 10, 13, 15, 20, 21, 22 were found to change significantly on ly in FR treated group but not in LPS treated group or control group. Interestin gly, the volume of xPr 13, 20, 21, 22 were increased and the volume of xPr 10, 1 5 were decreased. Conclusion: The molecular basis of thera peutic effect of FR on endotoxemia might be through the regulation of xPr1, 6, 7 , 8, 9, 11, 12, 14, 18, 23. We can use proteomic techniques to study the molecul ar mechanisms of diseases treated by functional Chinese herbs and the combinatio n of different herbs is necessary for the treatment of endotoxemia, as FR can no t regulated all the changed proteins induced by LPS.[
