红花药材的质量评价Quality evaluation of Flos Carthami

摘要(Abstract):
目的:对红花中色素类成分、腺苷及黄酮类化合物等成分进行分析,建立红花药材质量评价方法。方法:采用高效液相色谱-电化学检测技术,对和指纹图谱红花中羟基红花黄色素A的含量进行研究:Agilent ZORBAXSb C18色谱柱(4.6 mm×250 mm,5μm),以磷酸盐缓冲液-乙腈为流动相进行梯度洗脱,流速1.0 mL.min-1,柱温35℃,参比电极ISAAC(in-situ Ag/AgCl),工作电极为玻碳电极;辅助电极为铂电极;检测电位+800 mV。采用高效液相色谱-紫外检测技术,分析红花中腺苷的含量:Diamonsil C18色谱柱(4.6 mm×250 mm,5μm),流动相水-乙腈(95∶5),流速1.0 mL.min-1,柱温40℃,检测波长260 nm;采用紫外分光光度法,测定红花红色素的吸光度。结果:以21个共有峰为指标,建立了红花高效液相色谱指纹图谱,鉴定出7个色谱峰。32个红花样品中羟基红花黄色素A的质量分数为0.35%~3.58%;腺苷的质量分数为0.03‰~0.49‰。结论:该方法可用于红花质量评价。
Objective: To develop methods for qualitative and quantitative analyses of Flos Cartnami from three aspects,pigments,flavonoids and adenosine.Method: A method using HPLC coupled with electrochemical detector was developed to determine the content of hydroxysafflor yellow A and fingerprint of Flos Carthami.The chromatographic separation was performed on a Zorbax SB C18 column(4.6 mm×250 mm,5 μm) by gradient elution with phosphate buffer and acetonitrile at a flow-rate of 1.0 mL·min-1,the column temperature was 35 ℃,the reference electrode was ISAAC(in-situ silver/silver chloride),the working electrode was glassy carbon,the counter electrode was Pt,and the applied potential was+800 mV.Concentration of adenosine was determined by HPLC-UV on an Diamonsil C18 column(4.6 mm×250 mm,5 μm) with water-acetonitrile(95∶5) as mobile phase,the flow rate was 1.0 mL·min-1,the column temperature was 40 ℃ and the detection wavelength was 260 nm.The content of cartharmin was detected using a spectrophotometric method.Result: Twenty-one common chromatographic peaks were selected as characteristic peaks in the chromatogram of sample solution of Flos Cartnami.Seven peaks were identified as hydroxysafflor yellow A,6-hydroxykaempferol-3-O-glucoside,rutin,quercetin-3-O-glucoside,kaempferol-3-O-rutinoside,quercetin,kaempferol.The contents of hydroxysafflor yellow A and adenosine were from 0.35% to 3.58% and from 0.03‰ to 0.49‰,respectively.Conclusion: The methods can be used to evaluate the quality of Flos Carthami.
