熟地黄多糖对血虚模型小鼠的影响

摘要(Abstract):

目的 :观察熟地黄多糖 (RGP)对模型小鼠外周血象、骨髓造血干细胞、骨髓有核细胞的影响。方法 :各组小鼠连续灌胃 8~ 9d ,腹腔注射环磷酰胺 (CY)致化学损伤血虚模型 ,取全血测外周血象、骨髓有核细胞计数 ;各组小鼠尾静脉注射同种供体小鼠骨髓细胞悬液 ,取脾脏固定 ,观察外源性脾结节形成 ,各组小鼠以60 Coγ全身照射7 2 5Gy ,取脾脏观察内源性脾结节形成 ;各组小鼠60 Coγ全身照射 7 5Gy ,复制出放射性损伤模型 ,测外周血象和骨髓有核细胞数量。结果 :RGP各组对CY血虚模型小鼠外周血象、骨髓有核细胞下降均有拮抗作用 ,与模型组比较 ,有显著差异。对小鼠脾结节的形成有明显的促进作用 (P <0 0 5 ,P <0 0 1) ,对于小鼠外周血象的放射性损伤 ,RGP高剂量组可明显抑制小鼠全血细胞减少 (P <0 0 5 ) ,RGP低剂量组虽可提高损伤小鼠血细胞数量 ,但无统计学意义。结论 :熟地黄多糖对于不同血虚模型小鼠外周血象、骨髓有核细胞下降均有拮抗作用 ,对小鼠造血干细胞具有促进增殖、分化作用。
Objective: To observe the influence of RGP(Rehmanina glutinosa polyses) on the peripheral hemogram, hemopoietic stem cell,myeloid karyocyte of model mice Method: Each group of the mice were fed with RGP for 8~9 days Mice model of deficiency blood is set up by interaperitoneal injection of CY, and the peripheral hemogram and myeloid karyocyte were observed and counted on the whole blood Each group of mice were injected with the suspension of mycloid cell of homology donor mice, and the formation of splenic lymph nodules were observed Each group of mice were radiated with radiation dosage of 7 25GY, and the radiation damage model were set up The formation of splenic lymph nodules were observed, and the peripheral hemogram, myeloid karyocyte are observed and counted Result: Each dosage group of RGP have antagonism on peripheral hemogram and the myeloid karyocyte of the CY mice model of deficiency of blood, compared with control group RGP shows obvious facilitation anxo action for the formation of splenic lymph nodules (P<0 05, P<0 01) High dosage GRP can inhibition the decrease of whole blood cell obviously(P<0 05) Low dosage of RGP can increase the whole blood cell of modle mice without statistical meaning Conclusion: RGP have antagonism on the decrease of peripleral hemogram and myeloid karyocyte of different model mice of deficiency of blood, and have anxo action on multiplication and differentiation of the mice hemopoietic stem cell(CFu S)
