紫草素通过线粒体途径诱导A375-S2细胞凋亡

摘要(Abstract):

目的 :研究紫草素诱导人黑色素瘤A375 S2细胞凋亡的机制。方法 :MTT法测定紫草素对A375 S2细胞的生长抑制实验 ,形态学观察分析细胞凋亡的形态学变化 ,DNA电泳研究细胞死亡的机制 ,免疫印迹实验检测相关蛋白的表达。结果 :紫草素可明显地抑制A375 S2细胞的生长 ,在 2 5~ 4 0 μmol·L-1之间呈明显的量效关系和时效关系。 10 μmol·L-1紫草素可诱导A375 S2细胞凋亡 ,琼脂糖凝胶电泳可见典型的DNA“梯状”条道。caspase 3和caspase 9的抑制剂阻止紫草素诱导的凋亡 ,紫草素可诱导A375 S2细胞caspase 9的活力增加 ,免疫印迹结果显示的上调的Bax和下调的Bcl XL 表达证实了紫草素诱导的A375 S2细胞的凋亡是通过线粒体途径发挥作用的。结论 :10 μmol·L-1紫草素可明显地诱导A375 S2细胞凋亡 ,并经历了caspase 9激活的线粒体信号转导途径。
Objective: To study the mechanisms of shikonin induced A375 S2 cell apoptosis Method: Cytotoxicity assay by MTT, DNA fragmentation assay and Western blot analysis were carried out Result: 2 5~40 μmol·L -1 shikonin inhibited A375 S2 cell growth in a dose and time dependent manner 10 μmol·L -1 Shikonin induced apoptotic cell death in A375 S2 cells Caspase 3 inhibitor and caspase 9 inhibitor reversed shikonin induced A375 S2 cell apoptosis Up regulation of Bax and down regulation of Bcl X L experssion contributed to shikonin induced A375 S2 cell apoptosis Post mitochondrial caspase 9 dependent pathways participated in this apoptotic signal transduction pathway Conclusion: Mitochondria dependent pathways are involved in shikonin induced A375 S2 cell apoptosis
