广西菲牛蛭药材的高效毛细管电泳指纹图谱研究Study on HPCE fingerprints of Poecilobdella manillensis from Guangxi province

摘要(Abstract):
目的:采用高效毛细管电泳法建立广西菲牛蛭药材的指纹图谱。方法:石英毛细管柱(75μm×56 cm)作为分离通道,以25 mmol.L-1Na2HPO4-120 mmol.L-1Tris-16 mmol.L-1SDS(1 mol.L-1NaOH调pH 12.0)为缓冲液,运行电压17kV,柱温25℃,检测波长254 nm,进样参数3.4 kPa×6 s,运行时间27 min。结果:建立了广西菲牛蛭药材的高效毛细管电泳指纹图谱,确定了共有峰为13个,10批药材指纹图谱与对照指纹图谱相似度均大于0.98。结论:该方法具有良好的精密度、稳定性和重复性,为有效控制该药材质量提供了新方法。
Objective:To establish fingerprints for Poecilobdella manillensis from Guangxi province using the high performance capillary electrophoresis(HPCE) method.Method:Electrophoresis was performed on a fused silica capillary column(75 μm×56 cm),with 25 mmol·L-1 Na2HPO4-120 mmol·L-1 Tris-16 mmol·L-1 SDS(adjusted to pH 12.0 with 1 mol·L-1 NaOH) as the running buffer.The applied voltage was 17 kV,the temperature was 25 ℃ and the detection wavelength was 254 nm.The sample's hydrodynamic injection was 3.4 kPa×6 s and the duration was 27 min.Result:HPCE fingerprint was established with 13 common peaks.The similarity between fingerprints of P.manillensis in 10 batches and control fingerprints was more than 0.98.Conclusion:The method is so precise,reproducible and stable that it could be used as a new means for the quality control of P.manillensis.
