白芷中香豆素类成分的超临界流体萃取和GC-MS分析
Extraction of coumarins from Angelica dahurica by supercritical extraction and GC-MS analysis

摘要(Abstract):

目的 :对超临界CO2 萃取分离白芷中香豆素类成分进行研究。方法 :采用正交试验设计方法 ,以提取液中总香豆素含量为考察指标 ,对总香豆素提取条件进行优化 ,并用气相色谱 质谱联用技术对超临界样品进行分析鉴定 ,利用面积归一法测定其相对含量。结果 :气相色谱 质谱联用技术共鉴定出 15种香豆素类成分 ,主要成分氧化前胡素、欧前胡素、异欧前胡素的相对含量分别达到 42 .40 % ,2 2 .14 %和 12 .12 % ;最优萃取工艺为萃取压力 2 1MPa ,萃取温度 50℃ ,萃取时间 3 .0h ,颗粒度 2 0目 ,分离压力 6.5MPa ,分离温度 3 0℃。结论 :萃取压力、萃取温度、萃取时间、颗粒度、分离压力对实验指标有非常显著性影响
Objective: To extract coumarins from Ang el ica Dahurica by supercritical CO 2. Method: Six-factor and three-level orthogonal experimental design was used to optimize the SFE co nditions as guided by the content of total coumarins in the extract. Constituent s of the coumarins were analyzed by GC-MS, and their relative contents were det ermined by normalization method of areas. Result: Total 15 coumarins were separated, the relative contents of oxyimperatorin, imperatorin and isoimperatorin being up to 42.40%, 22.14% and 12.12% respectively. Optimum e xtraction process was established: 21 MPa as extraction pressure, 50 ℃ as extr action temperature, 3 h as time for extraction, 20 mesh as pulverized degree, 6 .5 MPa as separation pressure and 30 ℃ as separation temperature. Co nclusion: Changes in extraction pressure, temperature, time, pulveriz ed degree and separation pressure affect the extracting results remarkably.
