梁金菇多糖诱导K562细胞凋亡的研究

摘要(Abstract):

目的 :探讨梁金菇多糖对K5 6 2细胞凋亡的影响及其机制。方法 :应用集落形成实验观察梁金菇多糖对K5 6 2细胞增殖的影响 ,细胞形态学、DNA凝胶电泳等检测细胞凋亡 ,逆转录 聚合酶链反应 (RT PCR)检测凋亡相关基因FasmRNA的表达。结果 ;梁金菇多糖能明显抑制K5 6 2细胞的增殖 ,促进细胞凋亡 ,细胞凋亡率的增加呈剂量依赖性。Fas基因的表达与K5 6 2细胞凋亡呈正相关。结论 :梁金菇多糖具有抑制K5 6 2细胞增殖、诱导其凋亡的作用 ,Fas基因表达的上调可能是梁金菇多糖诱导细胞凋亡的机制之一。
Objective: To explore the effect of Liangjin mushroom polysaccharide on the apoptosis of K562 cells Method: The proliferation effect of Liangjin mushroom polysaccharide on K562 cells was observed by clone formating test, Apoptosis was detected by cyto morphology and DNA agarose gel electrophoresis, Expression of apoptosis related gene Fas mRNA was checked by RT PCR Result: Liangjin mushroom polysaccharide could significantly inhibit proliferation and promote apoptosis of K562 cells The apoptosis rate increased in a dose dependent manner There was a positive correlation between the expression of Fas and apoptosis of K562 cells Conclusion: Liangjin mushroom polysaccharide can inhibit proliferation and induce apoptosis of K562 cells The upregulation of expression of Fas gene may be one of the apoptotic mechanism
