摘要(Abstract):
该文建立了一种快速鉴别桂枝茯苓胶囊中苦杏仁苷、丹皮酚、芍药苷、肉桂酸、没食子酸以及苯甲酸的方法,该方法采用直接实时分析(DART)离子源串联四级杆-飞行时间质谱(Q-TOF),DART离子源以氦气为载气,温度450℃,样品传输速度0.2 mm·s-1;Agilent 6538 Q-TOF质谱毛细管电压1 000 V,干燥气温度350℃,气流速3.5 L·min-1,扫描模式为Target MS/MS扫描,扫描范围m/z 50~1 000。通过对照品与样品的一级、二级质谱图对比,快速鉴别出3批桂枝茯苓胶囊中的6种化学成分,该方法样品前处理简单,准确,快速,专属性强,能够广泛应用于中药复杂体系多成分的快速鉴别。
In order to establish a rapid method for identifying six constituents in Guizhi Fuling capsule,Q-TOF with DART ion source was used to perform the direct analysis of compounds in Guizhi Fuling capsule. The DART sampler delivery rate was 0. 2 mm·s- 1. The temperature of helium gas of DART was 450 ℃. The capillary voltage was kept at 1 000 V. The temperature of the drying gas of Agilent 6538 Q-TOF MS was set at 350 ℃. The flow rate of the drying gas of MS was set at 3. 5 L·min- 1. The MS scan range was m / z 50-1 000. Based on accurate mass measurements and the elemental compositions of the product ions and fragmentation patterns of reference conpounds,six components,amygdalin,paeonol,paeoniflorin,cinnamic acids,gallic acid,benzoic acid were identified rapidly. The method can rapidly identify six chemical constituents in three batch of Guizhi Fuling capsule. The DART-Q-TOF-MS method is simple,rapid and specific and it can be used for rapid identification and characterization of compounds in traditional Chinese medicines.
