水杉总黄酮对大鼠脑缺血再灌注损伤的保护作用

摘要(Abstract):

目的 :观察水杉总黄酮对大鼠脑缺血再灌注损伤的保护作用。方法 :凝闭大鼠双侧椎动脉 4~ 5h后 ,夹闭颈总动脉 30min ,再灌注 90min ,造成大鼠脑缺血再灌注损伤模型 ,观察水杉总黄酮对脑组织中生化指标及脑电图的影响。结果 :缺血再灌注造成脑组织严重损伤 ,脑组织中水 ,Na+ ,Ca2 + 含量及丙二醛 (MDA)含量较对照组明显增高 ,而超氧化物歧化酶 (SOD)活力较对照组降低 ,并伴有脑电图异常和脑组织结构病理性改变。夹闭颈总动脉前 30minip 2 5~ 10 0mg·kg-1水杉总黄酮能降低脑组织中水 ,Na+ ,Ca2 + 含量及MDA含量 ,并提高SOD活力 ,对组织学检查及脑电图也有所改善。结论 :水杉总黄酮对脑缺血再灌注损伤具有保护作用。
Objective: To observe the protective effects of total flavones of metasequosia(TFM) on cerebral ischemia-reperfusion injury in rats. Method: The bilateral vertebral arteries of rats were occluded under anesthesia,and 4~5 h later the carotid arteries of rats in the conscious condition were occluded for 30 min followed by reperfusion for 90 min. The effects of TFM on the contents of water,Na +,Ca 2+ in cortex,malondialdehyde (MDA) content and superoxide dismutase (SOD) activity in brain hemispheres,as well as the EEG activities were observed. Result: After 30 min ischemia and 90 min reperfusion,the contents of water,Na +,Ca 2+ and MDA were increased,and the SOD activity was reduced with abnormal EEG activity and ischemic injury in the brain tissues. TFM 25~100 mg·kg -1 ip 30 min before the carotid arteries were occluded,decreased the elevated water,Na +,Ca 2+ and MDA contents,increased the SOD activity ,reduced the ischemic injury of brain tissue,and promoted the recovery of EEG activities. Conclusion: TFM has a protective effect on cerebral ischemia-reperfusion injury in rats.
