HPLC-DAD-ELSD指纹图谱的化学模式识别用于黄芪质量评价

摘要(Abstract):

目的:建立黄芪药材的HPLC-DAD-ELSD色谱指纹图谱,采用相似度计算和化学模式识别的方法评价黄芪药材质量。方法:采用HPLC-DAD-ELSD串联技术,以Agilent Zorbax ODS C18色谱柱(4.6 mm×250 mm,5μm),乙腈-0.2%甲酸溶液为流动相进行梯度洗脱,流速0.8 mL·min-1,柱温25℃,进样量10μL,检测波长280 nm;ELSD汽化室温度110℃,氮气流速3.0 L·min-1。并对黄芪药材指纹图谱进行相似度计算和化学模式识别分析。结果:黄芪药材色谱指纹图谱测定方法稳定性、精密度和重复性良好,以18批黄芪药材建立指纹图谱共有模式,黄芪药材相似度均达到0.83,模式识别分析结果与其一致。结论:指纹图谱结合化学模式识别分析技术可以有效区分黄芪药材及其相关样品,可用于黄芪药材的质量评价,为药材的全面质量控制提供科学依据。
Objective: To develop an HPLC-DAD-ELSD method for detecting the fingerprint of Astragali Radix and evaluate the quality through similarity calculation and chemical pattern recognition. Method: Separation was performed at 25 ℃ on an Agilent Zorbax ODS C18column( 4. 6 mm × 250 mm,5 μm). Gradient elution was performed with the mobile phases of acetonitrile and water containing 0. 2% formic acid. The flow rate was 0. 8 mL· min- 1,and sample size was 10 μL. The UV detection wavelength was set at 280 nm. The drift tube temperature for ELSD was set at 110 ℃,and the nebulizing gas flow rate was 3. 0 L· min- 1. The similarity calculation and chemical pattern recognition were used for fingerprint analysis. Result: The HPLC-DAD-ELSD method for chromatographic fingerprint of Astragali Radix showed better results of stability,precision and repeatability. The reference chromatographic fingerprint of Astragali Radix was established on the eighteen Astragali Radix samples from different sources. The results of similarity calculation were higher than 0. 83,which was in accordance with the result of chemical pattern recognition analysis. Conclusion: Fingerprint and chemical pattern recognition analysis could effectively distinguish Astragali Radix from different source,which could be applied to the quality control of Astragali Radix.
