高效毛细管电泳法测定中药大黄及青海野生大黄茶中活性蒽醌类成分的含量

摘要(Abstract):

目的 :首次建立一种简单、快速且能同时分离测定中药大黄及青海野生大黄茶中 3种主要活性蒽醌成分含量的胶束毛细管电泳新方法。方法 :熔融石英毛细管柱 (75 μm× 5 0 .0cm ,有效长度 4 2 .4cm) ;缓冲体系 15 .0mmol·L-1硼砂 30 .0mmol·L-1SDS 10 %乙醇 ,pH 9.6 0 ,分离电压 2 0kV ;检测波长 2 5 4nm。结果 :大黄素、芦荟大黄素和大黄酸的线性范围分别为 4~ 12 0 (r=0 .992 1) ,10~ 2 0 0 (r=0 .9970 ) ,2~ 10 0mg·L-1(r=0 .9971)。日内迁移时间和峰面积的相对标准偏差分别为 0 .5 9%~ 0 .80 % ,1.30 %~ 3.2 2 % ,回收率范围为 97.6 %~ 10 2 .3%。结论 :方法简单、快速、准确、重现性好 ,可用于大黄药材和青海野生大黄茶的质量控制。
Objective: To establish an instant determi na tion method of emodin, aloe-emodin and rhein, from Rheum, and one of their prep arations, Qinghai Wild Dahuang Tea, by micellar electrokinetic capillary electro phoresis for the first time. Method: Separation was carrie d out in an uncoated fused silica capillary (75 μm×50.0 cm). Meanwhile, a runni ng voltage 20 kV, 15.0 mmol·L -1 borax buffer with 30.0 mmol·L -1 SD S and 10% ethanol (pH 9.60) and a UV detector at 254 nm were adopted. Result: The linear calibration rang was 4~120 mg·L -1 (r =0.992 1) for emodin, 10~200 mg·L -1 (r=0.997 0) for aloe-emodin, an d 2~100 mg·L -1 (r=0.997 1) for rhein, respectively. Under the optimu m conditions, the relative standard deviation (RSD) values (n=6) for the mig ration time and the peak area of each peak were 0.59%~0.80%, 1.30%~3.22%,resp ectively. The contents of the analytes were easily determined with recoveries ra nging from 97.6%~102.3%. Conclusion: The method is proved to be simple, rapid and accurate, and can be used for the quality control of me dicinal herb, Rheum, and its tea preparation.[
