GC-MS建立石菖蒲挥发油特征指纹图谱方法学研究

摘要(Abstract):

目的 :建立石菖蒲挥发油GC MS分析色谱条件 ,初步拟定挥发油指纹特征图谱指标成分群。方法 :采用GC MS法分析 10批石菖蒲挥发油化学成分。结果 :在进样口温度 2 5 0℃ ,接口温度 2 30℃ ,载气流速 1.3mL·min-1,柱压 80kPa ,电离电压 1.4kV ,升温速率 3℃·min-1的色谱条件下 ,石菖蒲挥发油主成分可达到较好分离 ;挥发油主要含有甲基丁香酚 (2 .13% )、顺式甲基异丁香酚 (4 .4 8% )、反式甲基异丁香酚 (0 .82 % )、γ 细辛醚 (4 .5 1% )、β 细辛醚 (6 6 .15 % )、α 细辛醚 (6 .35 % )等 6个特征成分 ;以此 6个主要共有峰为评价指标 ,GC MS色谱条件的精密度、稳定性、重现性良好。结论 :方法准确可靠 ,重复性好 ,可用于石菖蒲挥发油指纹图谱的研究
Objective: To establish the method of fingerprint analysis on volatile oil in rhizome of Acorus tatarinowii by GC-MS, and to study the main characteristic components. Method: The main components of 10 samples were determined by GC-MS. Result: The injector temperature was 250 ℃. The interface temperature was 230 ℃. The column flow was 1.3 mL·min~(-1). The column pressure was 80 kPa. The detector volt was 1.4 kV. The temperature rate was 3 ℃·min~(-1). And the main characteristic components were composed of the methyleugenol (2.13%), cis-methylisoeugenol (4.48%), trans-methylisoeugenol (0.82%), γ-asarone(4.51%), β-asarone(66.15%), α-asarone(6.35%). And the RSD of precision and reproducibility and stability was almost in the range of 5%. Conclusion: The method is reliable, accurate and can be used for fingerprint analysis of volatile oil of Acorus tatarinowii.
