葛根素对大鼠心肌细胞L型钙离子通道的影响

摘要(Abstract):

目的 :观察葛根素对大鼠L型钙离子通道的作用。方法 :恒流Langendorff灌流 ,I型胶原酶解分离 ,全细胞膜片钳技术记录离子电流。结果 :2 .4mmol·L- 1葛根素可以抑制单个大鼠心室肌细胞L型钙离子通道电流 ,且成时间依赖性 ;葛根素可以促进L型钙通道电流 电压 (I V)曲线的上移。结论 :葛根素可以抑制大鼠心室肌细胞的L型钙离子通道电流 ,并提示葛根素的抗心肌缺血和抗心律失常作用可能与抑制L型钙离子通道电流有关
Objective:To observe the effect of Puerari n on L-type calcium channel in isolated rat ventricular myocytes. Meth od: The cardiac ventricular myocytes were isolated enzymatically by L angendorff perfusion techniques at constant flow rate. Whole-cell recording of patch-clamp techniques was used to observe the current of L-type calcium chann el.Result: Puerarin 2.4 mmol·L -1 could inhibit the current of L-type calcium channel of rat ventricular myocytes and this inhibiti on was time-dependent. Purerarin elevated the current-voltage (I-V) curve of calcium current.Conclution:Puerarin can inhibit L-type ca lcium current of rat ventricular myocytes. Which implies that puerarin takes par t in anti-myocardial ischemia and anti-arrhythmics partly due to the inhibitio n of L-type calcium channel.
